BBL™ CHROMagar™ Staph aureus medium is designed for the isolation and identification of Staphylococcus aureus without the use of confirmatory testing from clinical sources. BBL CHROMagar Staph aureus medium can be utilized for the isolation and identification of S. aureus from food and environmental sources. BBL CHROMagar Staph aureus is highly effective in differentiating organisms with atypical appearance or weak hemolysis making it an ideal medium for Staphylococcus aureus surveillance.

Results
After proper incubation, read plates against a white background. S. aureus produces mauve to orange/mauve colored colonies and Staph saprophyticus appear green to blue-green on the CHROMagar medium. Most gram-positive organisms, if not inhibited, will produce white, blue, green or blue-green (aqua) colonies. Gram-negative organisms and yeasts are partially to completely suppressed.
Principles
Specially selected peptones supply the nutrients in BBL CHROMagar Staph aureus medium. The chromogen mix consists of artificial substrates (chromogens), which release an insoluble colored compound when hydrolyzed by specific enzymes. Selective agents have also been added for the suppression of gram-negative organisms and partial suppression of yeast.
Procedure
1. Enrich sample in enrichment broth i.e. peptone buffered broth at 35 degrees C. for 24 hours.
2. Inoculate CHROMagarTM Staph plate with 0.1ml of the enrichment broth and streak for isolation. Incubate at 35 degrees C. for 24 hours. If the specimen is cultured from a swab, roll the swab over a small area and streak from this area with a loop.
3. Observe plates for characteristic colonial morphology and color at 18-24 hours.
